为了解Ⅵ型胶原蛋白在人卵巢卵泡的生长过程中的表达,用鼠抗人Ⅵ型胶原蛋白特异性单克隆抗体,做免疫组化测定Ⅵ型胶原蛋白在卵泡生长过程中的分布特点,用原位杂交法检测卵泡组织中Ⅵ型胶原蛋白的mRNA表达。 人卵巢组织(包括卵泡)取自月经规则因子宫颈癌需做子宫附件切除的病人,年龄39~46岁,均未接受激素治疗,术前也无卵巢功能或内分泌紊乱。取材在月经周期中的具体时间判定根据月经史及基础体温测定。周期为28~30 d。卵泡期分为早卵泡期(周期第1~5 d)4例;中卵泡期(周期第6~9 d)3例;排卵前期(周期第10~14 d)5例。卵巢组织中未发现癌转移灶。 结果:免疫组化染色发现,取自早卵泡期的原始卵泡和初级卵泡未呈现Ⅵ型胶原蛋白阳性染色。中卵泡期次级卵
To understand the expression of collagen type Ⅵ in the growth of human ovarian follicles, the distribution characteristics of type Ⅵ collagen during follicular growth were determined by immunohistochemistry using mouse anti-human Ⅵ-type collagen-specific monoclonal antibody. Results: Immunohistochemical staining showed that the primordial follicles from the early follicular phase and the primary follicles did not show type Ⅵ collagen-positive staining.
