[目的]筛选肠道干细胞特征的差异蛋白质或蛋白质群。[方法]采用双向凝胶电泳方法分离新生小鼠肠边缘群细胞与肠上皮细胞总蛋白,选择差异表达超过1.5倍的蛋白质进行质谱分析及数据库搜索、鉴定。[结果]成功建立小鼠肠边缘群细胞与肠上皮细胞的双向电泳图谱,筛选出差异蛋白点39个,质谱分析及数据库搜索共鉴定出32种蛋白质。[结论]小鼠肠干细胞与上皮细胞的蛋白表达谱存在差异,鉴定出的32种蛋白质有可能成为潜在的、有应用价值的肠道干细胞鉴别分子标志。
[Objective] To screen differentiated proteins or protein groups characterized by intestinal stem cells. [Method] The total protein of intestinal border cells and intestinal epithelial cells of newborn mice was separated by two - dimensional gel electrophoresis. The proteins with more than 1.5 times difference were selected for mass spectrometry analysis and database search and identification. [Result] The two-dimensional electrophoresis pattern of mouse marginal swarm cells and intestinal epithelial cells was successfully established, and 39 differentially expressed protein spots were screened out. A total of 32 proteins were identified by mass spectrometry and database search. [Conclusion] The protein expression profiles of mouse intestinal stem cells and epithelial cells are different. The 32 proteins identified may be potential molecular markers of intestinal stem cell identification.
