目的:建立HPLC-ELSD同时测定咳灵胶囊中的苦杏仁苷、桃叶珊瑚苷、哈帕苷、贝母辛、贝母素甲和贝母素乙含量的方法。方法:采用Ultimate XB C_(18)色谱柱(250 mm×4.6 mm,5μm),蒸发光散射检测器(漂移管温度105℃,氮气流速2.0 L·min~(-1));以乙腈-甲醇(1∶1)与0.4%醋酸溶液为流动相,进行梯度洗脱,流速:0.7 ml·min~(-1),柱温:35℃。结果:苦杏仁苷、桃叶珊瑚苷、哈帕苷、贝母辛、贝母素甲和贝母素乙的线性范围分别为13.56~271.20μg·ml~(-1)(r=0.999 2)、8.48~169.60μg·ml~(-1)(r=0.999 9)、4.89~97.80μg·ml~(-1)(r=0.999 7)、2.66~53.20μg·ml~(-1)(r=0.999 4)、1.82~36.40μg·ml~(-1)(r=0.999 8)、2.04~40.80μg·ml~(-1)(r=0.999 6);平均加样回收率(RSD)分别为97.90%(1.20%),99.21%(1.62%),97.68%(0.75%),98.36%(1.38%),99.70%(0.79%),97.95%(1.56%)(n=6)。结论:本文建立的方法结果准确,重复性好,样品处理简便,可作为咳灵胶囊的质量控制方法。
OBJECTIVE: To establish a HPLC-ELSD method for the simultaneous determination of amygdalin, aucubin, harpagoside, fumonisin, peiminine and peiminine B in Kelin capsules. Methods: The final XB C_ (18) column (250 mm × 4.6 mm, 5 μm) was used to evaporate the light scattering detector (drift tube temperature was 105 ℃ and nitrogen flow rate was 2.0 L · min -1) (1: 1) and 0.4% acetic acid as mobile phase, gradient elution was carried out at a flow rate of 0.7 ml · min -1. The column temperature was 35 ℃. RESULTS: The linear ranges of amygdalin, aucubin, harpagoside, fosfacilim, peginterferon and peiminine were 13.56-2.71.20μg · ml -1 (r = 0.999 2) , 8.48 ~ 169.60μg · ml -1 (r = 0.999 9), 4.89-97.80μg · ml -1 (r = 0.999 7), 2.66 ~ 53.20μg · ml -1 (r = 0.999 4), 1.82- 36.40μg · ml -1 (r = 0.999 8) and 2.04-40.80μg · ml -1 (r = 0.999 6) respectively. The average recovery rates Were 97.90% (1.20%), 99.21% (1.62%), 97.68% (0.75%), 98.36% (1.38%), 99.70% (0.79%), 97.95% (1.56%) (n = 6). Conclusion: The method established in this paper is accurate, reproducible and simple to handle. It can be used as a quality control method for Kelin capsule.
