目的筛选甘草的醇沉淀物经柱层析后具有体外抗呼吸道合胞病毒(RSV)感染作用的洗脱部分。方法采用水提醇沉法制出甘草的醇沉物,经大孔吸附树脂HPD722、HPD750、D101柱层析、蒸馏水洗脱出多个部位;以利巴韦林为阳性对照药物,采用MTT法检测洗脱部分与甘草酸在Hep-2细胞中的药物毒性及抗RSV效果。结果 HPD722水部位、HPD750水部位、D101水部位、甘草酸、利巴韦林对RSV感染的治疗指数(TI)依次为13.698、261.578、1.461、1.708、27.895。结论甘草醇沉物在体外对RSV具有显著的抑制作用,HPD750水部位可能是其主要的有效部位,值得进一步深入研究。
Objective To screen the elution fractions of glycogen alcoholic precipitate after column chromatography with anti-respiratory syncytial virus (RSV) infection in vitro. Methods Alcohol precipitates of liquorice were prepared by water extraction and alcohol precipitation. The fractions were eluted by macroporous resin HPD722, HPD750, D101 and distilled water. Ribavirin was used as a positive control drug and MTT assay Pharmacological toxicity and anti-RSV effect of elution fraction and glycyrrhizin in Hep-2 cells. Results HPD722 water, HPD750 water, D101 water, glycyrrhizin, ribavirin RSV infection treatment index (TI) were 13.698,261.578,1.461,1.708,27.895. Conclusion The glycyrrhizic alcohol precipitates have a significant inhibitory effect on RSV in vitro. The water fraction of HPD750 may be the main effective site, which deserves further study.
