目的建立高效毛细管电泳法(HPCE)同时测定不同批次玄参中梓醇、桃叶珊瑚苷、哈巴苷、哈巴俄苷、毛蕊花糖苷和肉桂酸含量的方法。方法以60 mmol.L-1硼砂缓冲液(pH 9.3)为电泳介质,未涂渍标准熔融石英毛细管(75μm×64.5 cm,有效长度56 cm)为分离通道,分离电压为20 kV,检测波长为210 nm,毛细管温度为25℃,压力进样为5 kPa×6 s。结果 6种指标成分的浓度与峰面积的线性关系良好(r>0.997 1);加样回收率为97.05%~103.43%。用此法测定了玄参药材中梓醇、桃叶珊瑚苷、哈巴苷、哈巴俄苷、毛蕊花糖苷和肉桂酸等6种指标成分的含量,得到满意结果。结论该方法简单、准确,重复性较好,可用于玄参药材内在质量的评价和控制。
OBJECTIVE To establish a high performance capillary electrophoresis (HPCE) method for the simultaneous determination of catalpol, aucubin, hababine, harpagoside, verbascoside and cinnamic acid in different batches of Scrophulariaceae. Methods 60 mmol·L-1 borax buffer (pH 9.3) was used as the electrophoresis medium. The standard fused silica capillary (75 μm × 64.5 cm, effective length 56 cm) was used as the separation channel. The separation voltage was 20 kV and the detection wavelength was 210 nm, capillary temperature 25 ℃, pressure injection 5 kPa × 6 s. Results The linear relationship between the concentration of 6 kinds of index components and the peak area was good (r> 0.997 1). The average recoveries were 97.05% -103.43%. The contents of 6 index components of catalpol, aucubin, habobin, harbutazone, abacus glycoside and cinnamic acid in Radix Astragali were determined by this method, and satisfactory results were obtained. Conclusion The method is simple, accurate and reproducible. It can be used to evaluate and control the inherent quality of Radix Scrophulariaceae.
