作者研究具不同溶解度参数的介质对睾酮经皮渗透的影响,测定了药物在介质中的溶解度及通过大鼠皮肤的渗透速率。经Hildbrand法计算得到不同介质的溶解度参数(δv(n)),并由热力学公式进一步计算出药物在不同介质与皮肤间的分配系数(k)。结果显示,当介质的溶解度参数在一定范围内时,实验测得的药物渗透系数(p)的对数与药物理论分配系数(k)的对数有良好的相关性。
The authors studied the effect of media with different solubility parameters on transdermal penetration of testosterone and determined the drug’s solubility in the medium and the rate of penetration through the rat’s skin. The solubility parameter (δv (n)) of different media was calculated by Hildbrand method, and the partition coefficient (k) of drug between different media and skin was further calculated by thermodynamic formula. The results show that when the solubility parameter of the medium is within a certain range, the experimental logarithm of the drug permeability coefficient (p) has a good correlation with the logarithm of the theoretical drug partition coefficient (k).
