[目的]研究没食子儿茶素没食子酸酯(EGCG)对乳腺癌MCF-7细胞生长及其HIF-1α和VEGF表达的影响。[方法]分别用浓度为25、50、100mg/L的EGCG处理人乳腺癌细胞株MCF-748h后,采用台盼蓝染色法检测各组细胞生长增殖情况;RT-PCR法检测各组细胞中HIF-1α和VEGF mRNA的表达;Western Blot检测各组细胞中HIF-1α蛋白的表达;ELISA法检测各条件培养液中VEGF蛋白的分泌水平。[结果]经不同浓度EGCG处理的乳腺癌细胞,其细胞生长速度明显变缓,且随着药物浓度的增加,生长变缓更明显(P=0.007);不同浓度的EGCG处理MCF-7细胞后,细胞中VEGF mRNA或蛋白表达量均有明显下降(P<0.05);不同浓度的EGCG处理MCF-7细胞后,细胞中HIF-1α蛋白的相对表达量下降(P<0.05)。[结论]EGCG可抑制乳腺癌细胞的生长增殖,可能与抑制HIF-1α和VEGF蛋白的表达有关。
[Objective] To study the effect of EGCG on the growth of breast cancer MCF-7 cells and the expression of HIF-1α and VEGF. [Methods] The human breast cancer cell line MCF-748h was treated with EGCG at a concentration of 25, 50 and 100mg / L, respectively. The growth and proliferation of each group were detected by trypan blue staining. HIF-1α and VEGF mRNA were detected by Western Blot. HIF-1αprotein expression was detected by Western Blot. The secretion of VEGF protein in each culture medium was detected by ELISA. [Result] The cell growth rate of the breast cancer cells treated with different concentrations of EGCG was significantly slowed down, and the growth became more obvious with the increase of the drug concentration (P = 0.007). After treated with different concentrations of EGCG, the growth of MCF- (P <0.05). The relative expression of HIF-1α protein in MCF-7 cells treated with different concentrations of EGCG decreased (P <0.05). [Conclusion] EGCG can inhibit the growth and proliferation of breast cancer cells, which may be related to the inhibition of the expression of HIF-1α and VEGF protein.
