目的研究NYGGF4(PID1)基因过表达对脂肪细胞中脂肪酸合成酶(FASN)的影响。方法体外培养稳定转染NYGGF4(PID1)基因的3T3-L1前体脂肪细胞株[NYGGF4-pcDNA3.1(+)/myc-His B,A组]及空载体对照细胞株[pcDNA3.1(+)/myc-His B,C组],经1-甲基-3异丁基黄嘌呤、地塞米松、胰岛素方案诱导分化为成熟脂肪细胞后,采用RT-PCR和Western blot法检测FASN基因mRNA和蛋白表达水平。结果与C组相比,A组成熟脂肪细胞FASN mRNA及蛋白表达水平均明显增高(P<0.05)。结论 NYGGF4(PID1)基因过表达可能影响成熟脂肪细胞中脂肪酸的合成。
Objective To investigate the effect of over-expression of NYGGF4 gene on fatty acid synthase (FASN) in adipocytes. Methods The 3T3-L1 precursor adipocyte strain [NYGGF4-pcDNA3.1 (+) / myc-His B, group A] stably transfected with NYGGF4 (PID1) gene and pcDNA3.1 ) / myc-His B, C group]. After differentiated into mature adipocytes by 1-methyl-3 isobutylxanthine, dexamethasone and insulin, the mRNA expression of FASN was detected by RT-PCR and Western blot And protein expression level. Results Compared with group C, the expression of FASN mRNA and protein in mature adipocytes of group A were significantly increased (P <0.05). Conclusion Overexpression of NYGGF4 (PID1) gene may affect fatty acid synthesis in mature adipocytes.
