目的:探讨非小细胞肺癌中DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶基因的过甲基化状况及表达的关系。方法:采用甲基化特异性PCR及免疫组织化学法检测96例经病理确诊的非小细胞肺癌中DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶基因及蛋白表达。结果:96例非小细胞肺癌组织DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶基因过甲基化率36.5%,DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶蛋白的阳性表达率为33.3%,DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶基因过甲基化与蛋白的表达呈负相关(Rs=-0.536,P<0.05)。结论:在非小细胞肺癌中,DNA修复酶O6-甲基鸟嘌呤-DNA甲基转移酶基因过甲基化导致蛋白表达的降低,可能与肺癌的发生、发展相关。
Objective: To investigate the hypermethylation status of DNA repair enzyme O6-methylguanine-DNA methyltransferase gene and its expression relationship in non-small cell lung cancer. Methods: The DNA repair enzyme O6-methylguanine-DNA methyltransferase gene and protein expression in 96 cases of non-small cell lung cancer confirmed by pathology were detected by methylation-specific PCR and immunohistochemistry. RESULTS: The methylation rate of DNA repair enzyme O6-methylguanine-DNA methyltransferase in 96 cases of non-small cell lung cancer was 36.5%, and DNA repair enzyme O6-methylguanine-DNA methyltransferase protein The positive rate was 33.3%. The hypermethylation of the DNA repair enzyme O6-methylguanine-DNA methyltransferase gene was negatively correlated with protein expression (Rs=-0.536, P<0.05). Conclusion: In non-small cell lung cancer, the hypermethylation of the DNA repair enzyme O6-methylguanine-DNA methyltransferase gene leads to the decrease of protein expression, which may be related to the occurrence and development of lung cancer.
