以茶薪菇SM菌株为试材,研究了4种分离纯化方法对厚垣孢子分离纯化的影响,并观察其显微萌发特征。结果表明:研磨酶解方法较好,在1.0%蜗牛酶和1.0%纤维素酶,30℃、4h条件下,菌丝碎片酶解消失,孢子饱满,数量达1.9×105个/mL;显微镜检发现多数厚垣孢子萌发点位于孢子侧面,萌发菌丝具锁状联合结构,萌发后原孢子的液泡体积增大,胞内充满大液泡。
The SM strains of Agrocybe boulardii were used as materials to study the effects of four isolation and purification methods on the isolation and purification of chlamydospores and to observe their micro-germination characteristics. The results showed that the method of grinding and enzymolysis was better. Under the conditions of 1.0% snail enzyme and 1.0% cellulase, the mycelial fragments disappeared and the number of spores was full, reaching 1.9 × 105 / mL under the conditions of 30 ℃ and 4h. The majority of chlamydospore germination sites were found on the side of the spores. The germinating mycelium had a lock-like structure. After the germination, the volume of vacuoles of the sporozoites increased and the intracellular vacuole filled.
