目的:建立一种可靠、实用的局部脑缺血再灌注模型。方法:用尼龙线栓塞大脑中动脉(MCA)制做局部脑缺血再灌注模型。观察大鼠脑缺血不同时间再灌注的成功率、神经经病学评分、丙二醛(MDA)含量、脑水肿及病理学变化。结果:MCA闭塞24h后,大鼠出现脑水肿、脂质过氧化和不同程度的神经损伤症状;光镜下见额顶叶皮质及基底节区梗塞。早期再灌注可明显改善神经损伤症状,减轻脑水肿,而脂质过氧化及病理损害加重。改进实验方法后,大鼠脑缺血不同时间再灌注的成功率明显提高。结论:该模型是一种较理想的脑缺血再灌注模型
Objective: To establish a reliable and practical model of focal cerebral ischemia-reperfusion. Methods: The model of focal cerebral ischemia-reperfusion was established by embolization of middle cerebral artery (MCA) with nylon thread. The success rate of reperfusion, neurological score, malondialdehyde (MDA) content, brain edema and pathological changes at different time points were observed. Results: After occlusion of MCA for 24 hours, rats developed cerebral edema, lipid peroxidation and different degrees of nerve injury symptoms. The frontal cortex and basal ganglia infarction were observed under light microscope. Early reperfusion can significantly improve the symptoms of nerve injury, reduce brain edema, and lipid peroxidation and pathological damage worse. After improving the experimental method, the success rate of reperfusion in rats with cerebral ischemia at different time was significantly improved. Conclusion: This model is a better model of cerebral ischemia-reperfusion
