【目的】本文为提高使君子氨酸的提取收率并探讨使君子粗提液的酶解工艺,研究了本实验室筛选的J6菌所产蛋白酶对使君子粗提液的酶解特性。【方法】将J6菌所产蛋白酶在不同时间、p H及温度情况下分析蛋白酶对使君子粗提液中使君子氨酸含量的影响。【结果】在使用蛋白酶为5000U/ml,温度为20℃,p H10.0,酶解时间为24小时的条件下酶解得到的使君子氨酸含量最高,能有效提稿游离使君子氨酸含量达20%。【结论】J6菌所产蛋白酶促进了使君子的水解,加速了使君子氨酸的游离,可望将该菌所产蛋白酶用于使君子氨酸的粗提工艺中。
【Objective】 In order to improve the yield of quercetin and to explore the enzymatic hydrolysis process of crude extract of Gentleman, the enzymolysis characteristics of crude extract from Geranium chinense were studied in this paper. 【Method】 The protease produced by J6 bacteria was analyzed at different times, pH and temperature. 【Result】 The results showed that the highest content of quisqualic acid was obtained when the protease was 5000U / ml, the temperature was 20 ℃, p H10.0 and the enzymolysis time was 24 hours. Content up to 20%. 【Conclusion】 The protease produced by J6 strain promoted the hydrolysis of the gentleman and accelerated the dissociation of quisqualic acid. It is expected that the protease produced by this strain will be used in crude extraction of quisqualic acid.
