目的 观察脂多糖 (LPS)诱导D -半乳糖 (D -gal)致衰老大鼠的急性肺损伤 (ALI)及氨基胍 (AG)对其是否有保护作用。方法 大鼠 2 4只随机分成两部分 :6只为正常对照组 ,18只经腹腔注射D -gal复制衰老动物模型。后者再随机分成 3组 :衰老对照组 (6只 )、LPS组 (6只 ,静脉注射LPS 5mg/kg,诱导形成ALI)、AG +LPS组 (6只 ,注射LPS前 30min静脉注射AG 2 5mg/kg)。注射LPS或生理盐水后 2h收集标本待测。结果 D -gal致衰老大鼠较正常对照组大鼠血中乳酸脱氢酶(LDH)活性显著升高 ,而肺组织中Na+ -K+ -ATPase活性下降 (均P <0 0 5 )。衰老大鼠注射LPS后 2h已形成ALI。肺泡灌洗液 (BALF)中蛋白含量增加 (P <0 0 1) ;BAIF中蛋白含量与血浆中蛋白含量比值 (PPI)升高 (P <0 0 1) ;肺湿干重量比增加(P <0 0 1)。血中乳酸 (LD)、丙二醛 (MDA)、一氧化氮 (NO)含量和乳酸脱氢酶 (LDH)活性 ,以及肺组织中髓过氧化物酶 (MPO)活性均显著升高 (均P <0 0 1) ;而肺组织中Na+ -K+ -ATPase活性显著下降 (P <0 0 5 )。预先给予AG可显著地缓解上述指标的变化 (P <0 0 5 ,P <0 0 1)。结论 D -gal致衰老大鼠机体已发生一定的退化性改变。静脉注射LPS可引起衰老大鼠明显的ALI。AG对LPS诱导衰老大鼠的ALI有显著的保护作用。
Aim To investigate whether acute lung injury (ALI) and aminoguanidine (AG) induced by lipopolysaccharide (LPS) induced aging in D-galactose rats have protective effects on it. Methods Twenty-four rats were randomly divided into two groups: six were normal control group and 18 were injected intraperitoneally with D -gal model. The latter were randomly divided into three groups: aged control group (6 rats), LPS group (6 rats injected with LPS 5mg / kg, induced ALI formation), AG + LPS group (6 rats injected AG 2 5 mg / kg). After injection of LPS or saline 2h specimens were tested. Results D-gal-induced aging rats had significantly higher levels of lactate dehydrogenase (LDH) activity and lower levels of Na + -K + -ATPase activity in lung tissue than those in normal control rats (all P <0.05). ALI was formed 2 hours after the injection of LPS into aged rats. The content of protein in BALF was increased (P <0.01), the protein content in BAIF was higher than that in plasma (P <0.01), and the weight ratio of lung wet to dry increased <0 0 1). Blood lactate (LD), malondialdehyde (MDA), nitric oxide (NO) and lactate dehydrogenase (LDH) activity, and myeloperoxidase (MPO) activity in lung tissue were significantly increased P <0.01). However, the activity of Na + -K + -ATPase in lung tissue was significantly decreased (P <0.05). Pretreatment with AG significantly ameliorated the above changes (P <0.05, P <0.01). Conclusion D-gal induced aging rats have some degenerative changes. Intravenous injection of LPS can cause significant ALI in aging rats. AG has a significant protective effect on ALI induced by LPS in aging rats.
