目的研究葛根素对人胆管癌QBC939细胞生长抑制的影响。方法首先体外培养人胆管癌QBC939细胞株,应用不同浓度葛根素处理QBC939细胞株,应用CCK8法检测不同浓度葛根素对胆管癌QBC939细胞的生长抑制作用;应用RT-PCR法检测葛根素作用于胆管癌QBC939细胞后细胞中血管内皮生长因子-C(VEGF-C)、环氧合酶-2(COX-2)基因表达变化。结果 (1)CCK8法检测结果显示,葛根素对胆管癌QBC939细胞的生长具有明显抑制作用,且随着浓度的增加,其对胆管癌QBC939细胞的抑制作用越明显;(2)RT-PCR法显示葛根素明显抑制胆管癌QBC939细胞中VEGF-C,COX-2基因表达。结论葛根素对人胆管癌QBC939细胞的生长有显著抑制作用。
Objective To study the effect of puerarin on the growth inhibition of human cholangiocarcinoma QBC939 cells. Methods QBC939 human cholangiocarcinoma cell line was cultured in vitro. QBC939 cells were treated with different concentrations of puerarin. CCK8 assay was used to detect the inhibitory effects of different concentrations of puerarin on the growth of human cholangiocarcinoma QBC939 cells. RT- The expression of vascular endothelial growth factor-C (VEGF-C) and cyclooxygenase-2 (COX-2) gene in QBC939 cells was detected by RT-PCR. Results (1) CCK8 assay showed that puerarin significantly inhibited the growth of human cholangiocarcinoma QBC939 cells. With the increase of concentration, puerarin inhibited the growth of cholangiocarcinoma QBC939 cells more easily. (2) RT-PCR Puerarin significantly inhibited VEGF-C, COX-2 gene expression in cholangiocarcinoma QBC939 cells. Conclusion Puerarin has a significant inhibitory effect on the growth of human cholangiocarcinoma QBC939 cells.
