目的研究帕金森病(PD)黑质多巴胺神经元的死亡机理。方法用脱氧核糖核酸(DNA)标记法,借助流式细胞仪和荧光显微镜观察多巴胺(DA)对大鼠嗜铬细胞瘤(PC12)细胞凋亡的影响。结果DA可诱发PC12细胞凋亡,适中浓度(0.45mmol/L)时PC12细胞凋亡数最多(凋亡率为48.7%±6.3%);抗氧化剂二硫苏糖醇(DTT)可部分阻止DA诱发的PC12细胞凋亡(P<0.01)。结论细胞凋亡参与了PD的病变过程,采用适当的抗氧化剂可能有益于PD的治疗
Aim To study the mechanism of death of dopaminergic neurons in Parkinson’s disease (PD). Methods The effect of dopamine (DA) on the apoptosis of pheochromocytoma (PC12) cells was observed by flow cytometry and fluorescence microscopy with DNA labeling. Results DA induced apoptosis in PC12 cells. The apoptotic rate of PC12 cells was the highest (48.7% ± 6.3%) at moderate concentration (0.45 mmol / L); the activity of DTT ) Could partially prevent the apoptosis of PC12 cells induced by DA (P <0.01). Conclusions Apoptosis is involved in the pathological changes of PD. Adopting suitable antioxidants may be beneficial to the treatment of PD
