采用微量DNA提取技术,从梅花鹿血、毛、鹿鞭、鹿茸、牛鞭、驴鞭中提取DNA,以线粒体DNA细胞色素b通用引物L14841和H15149扩增约307bpDNA片段,扩增产物纯化后采用双脱氧链终止法测定其序列。结果证明:梅花鹿毛、血和鹿鞭的DNA序列完全一致;而所谓的“鹿茸”则与其有较大的差异。用所测序列以简约法PAUP311程序构建的分子系统树与传统分类系统相吻合。说明此方法鉴定鹿鞭是科学而准确的。
A DNA extraction technique was used to extract DNA from sika deer blood, hair, deer whip, antler, bullwhip, and donkey whip. Approximately 307 bp DNA fragment was amplified from mitochondrial DNA cytochrome b universal primers L14841 and H15149. The amplified product was purified and doubled. The deoxygenation chain termination method was used to determine its sequence. The results showed that the DNA sequences of deer hair, blood and deer whip were exactly the same, while the so-called “antler” was quite different from it. The molecular phylogenetic tree constructed using the reduced sequence PAUP311 program with the measured sequences agrees with the traditional classification system. Explain that this method of identifying Lubian is scientific and accurate.
