Defatted fruit bodies of Hericium erinaceus were extracted with 1 mol/L NaOH to obtain alkali-soluble polysaccharide (ASP). Several chemical and physical properties of ASP have been evaluated based on specific optical rotation ([α]_ D ) values (SOR) and sugar composition. Fractional precipitation with methanol, anion-exchange chromatography and gel chromatography were used to isolate ASP sub-fractions and a purified alkali-soluble glucan preparation (ASG) with a SOR of +278° and a glucose content of 96.1%.
