目的探讨外周血单个核细胞(peripheral blood mononuclear cell,PBMNC)中核糖体蛋白(ribosonoal protein S 26,RPS 26)和卵巢颗粒细胞中过氧化物酶体增殖物激活受体γ(peroxisome proliferator-activated receptor gamma,PPARγ)基因表达与多囊卵巢综合征(polycystic ovary syndrome,PCOS)患者病情的相关性。方法将深圳市龙华新区人民医院妇科2015年7月至2016年3月诊治的38例PCOS患者作为PCOS组,同期体检健康女性30例作为对照组,抽取外周血,采用Ct值比较法,通过SYBR Green II实时定量PCR检测两组PBMNC中RPS 26及PPARγ基因在mRNA的表达水平,进一步分析PCOS患者两基因表达水平及与体质量指数(body mass index,BMI)、性激素水平相关性。结果 PCOS组RPS 26表达水平高于对照组,但差异无统计学意义(P>0.05),PCOS组PPARγ基因表达明显高于对照组,差异有统计学意义(P<0.05)。RPS 26及PPARγ基因表达水平与睾酮呈正相关(P<0.05),与促卵泡成熟素呈负相关(P<0.05)。结论 RPS 26及PPARγ基因在PCOS患者PBMNC中的表达均高于正常女性,可不同程度地反映疾病的严重程度。
Objective To investigate the expression of peroxisome proliferator-activated receptor (γ) in ribosonoal protein S 26 (RPS 26) and ovarian granulosa cells in peripheral blood mononuclear cells (PBMNC) gamma, PPARγ) gene expression in patients with polycystic ovary syndrome (PCOS). Methods 38 PCOS patients diagnosed and treated by gynecological department of Longhua New District People’s Hospital of Shenzhen from July 2015 to March 2016 were selected as the PCOS group and 30 healthy women in the same period as the control group. Peripheral blood was collected and compared by Ct value comparison method and SYBR Green II real-time PCR was used to detect the mRNA expression of RPS 26 and PPARγ in PBMNC of both groups. The expression of two genes in PCOS patients was further analyzed, and their correlations with body mass index (BMI) and sex hormone levels were also analyzed. Results The expression of RPS26 in PCOS group was higher than that in control group, but the difference was not statistically significant (P> 0.05). The expression of PPARγ gene in PCOS group was significantly higher than that in control group (P <0.05). RPS 26 and PPARγ mRNA expression levels were positively correlated with testosterone (P <0.05) and negatively correlated with follicle-stimulating hormone (P <0.05). Conclusion The expressions of RPS 26 and PPARγ in PBMNC of patients with PCOS are higher than those of normal women, which can reflect the severity of the disease to varying degrees.
