目的:观察何首乌主要成分二苯乙烯苷(THSG)对去卵巢大鼠衰老骨髓间充质干细胞(MSCs)成骨、成脂分化的影响。方法:使用雌性大鼠双侧卵巢切除增龄3月法复制衰老大鼠模型,用THSG给予部分模型大鼠灌胃。分离、培养正常组、模型组、THSG组大鼠MSCs。观察各组大鼠MSCs骨向分化、脂向分化诱导的不同表现。结果:骨向诱导18、21天时,模型组、THSG组MSCs碱性磷酸酶(ALP)表达量低于正常组(P<0.01,P<0.05)。14天开始,模型组、THSG组大鼠MSCs的ALP染色阳性率低于正常组(P<0.01,P<0.05)。脂向诱导18天时,模型组细胞上清液中甘油三酯(TG)表达量高于正常组、THSG组(P<0.05)。21天时,模型组脂肪细胞阳性率高于正常组、THSG组(P<0.05)。结论:去卵巢大鼠衰老MSCs骨向分化能力降低,脂向分化能力增强。THSG可抑制衰老MSCs的脂向分化。
Objective: To observe the effect of stilbene glycoside (THSG), a major component of Polygonum multiflorum Thunb. On osteoblastic and adipogenic differentiation of aging mesenchymal stem cells (MSCs) in ovariectomized rats. Methods: Aged rats were treated with bilateral ovariectomy in aged rats and the rats were sacrificed by THSG. Isolated, cultured normal group, model group, THSG group rats MSCs. Observe the different manifestations of osteogenic differentiation and adipogenic differentiation of MSCs in each group. RESULTS: At 18 and 21 days after induction of bone, the expression of alkaline phosphatase (ALP) in MSCs in model group and THSG group was lower than that in normal group (P <0.01, P <0.05). At 14 days, the positive rate of ALP staining in MSCs of model group and THSG group was lower than that of normal group (P <0.01, P <0.05). At 18 days of lipid induction, the expression of triglyceride (TG) in the supernatant of the model group was higher than that in the normal group and the THSG group (P <0.05). At 21 days, the positive rate of adipocytes in model group was higher than that in normal group and THSG group (P <0.05). Conclusion: The bone marrow mesenchymal stem cells of ovariectomized rats have the ability of differentiating into bone and enhancing the differentiation of adipose tissue. THSG inhibits the adipogenic differentiation of aged MSCs.
