目的 观察肿瘤抗原激活的树突状细胞 (dendriticcell,DC)增强结直肠癌患者抗肿瘤免疫功能 ,抑制术后复发和转移的可能性。方法 分离结直肠癌根治术后患者外周血单个核细胞 (pe ripheralbloodmonocytecell,PBMC) ,以人大肠癌细胞系LOVO细胞抗原粗提物激发PBMC中DC ,以粒 /巨噬细胞集落刺激因子 (granulocyte/macrophagecolonystimulatingfactor,GM CSF)和白介素 4(interleukin4,IL 4)联合刺激产生DC ,将DC回输给结直肠癌患者 ,检测结直肠癌患者外周血癌胚抗原 (carcinoem bryonicantigen ,CEA)含量。结果 术后 12个月 ,实验组 12例患者的CEA含量为 (3± 1 4)ng/ml,未发现结直肠癌复发和转移 ;对照组 13例外周血CEA含量为 (39± 2 2 )ng/ml,其中 5例外周血CEA含量较高的患者〔(99± 47)ng/ml〕中 3例发生结直肠癌转移和 (或 )复发 (2例肝转移 ,1例复发并肝转移 )。结论 肿瘤抗原激活的DC能够增强结直肠癌患者抗肿瘤免疫功能 ,抑制术后结直肠癌的复发和转移
Objective To observe the effect of tumor antigen-activated dendritic cells (DCs) on the anti-tumor immune function of patients with colorectal cancer and to suppress the possibility of postoperative recurrence and metastasis. Methods Peripheral blood mononuclear cells (PBMC) were isolated from patients after radical resection of colorectal cancer. DCs of PBMCs were stimulated with crude extracts of LOVO cell antigen from human colorectal cancer cell line. Granulocyte/macrophage colony-stimulating factor (granulocyte/ Macrophage colony stimulating factor (GM CSF) and interleukin 4 (interleukin4, IL 4) are co-stimulated to produce DC, and DCs are returned to patients with colorectal cancer to detect the content of carcinoem bryonic antigen (CEA) in peripheral blood of patients with colorectal cancer. Results After 12 months of operation, the CEA content of the 12 patients in the experimental group was (3±14) ng/ml, and no recurrence or metastasis of colorectal cancer was found. The CEA content in the peripheral blood of 13 controls was (39±2 2) in the control group. Ng/ml, of which 5 patients with high weekly CEA content ((99 ± 47) ng/ml) had colorectal cancer metastasis and/or recurrence (2 liver metastases, 1 recurrence, and liver metastases) ). Conclusion Tumor antigen-activated DC can enhance the anti-tumor immune function of patients with colorectal cancer, and inhibit the recurrence and metastasis of colorectal cancer after operation.
