目的了解驱铅治疗后铅中毒患者外周血中T细胞受体信号转导分子CD3γ、CD3δ、CD3ε链表达情况。方法采用实时荧光定量PCR分别检测9例健康人及16例铅中毒患者临床上进行驱铅治疗前后外周血单个核细胞中CD3γ、CD3δ、CD3ε链mRNA水平,以β2微球蛋白(β2M)基因为内参基因,根据相对定量公式2-△Ct×100%计算各基因表达水平。采用Spearman秩相关分析健康人和铅中毒患者驱铅治疗前后CD3γ、CD3δ、CD3ε链mRNA水平相关性。结果铅中毒患者驱铅治疗前后CD3γ链mRNA水平与健康人相比无明显差异;驱铅治疗前后CD3δ链mRNA水平均显著低于健康人,驱铅治疗后CD3δ链mRNA水平较治疗前更低;驱铅治疗前CD3ε链mRNA水平与健康人相比显著升高,驱铅治疗后CD3ε链mRNA水平有所恢复。健康人中CD3γ链与CD3δ链、CD3γ链与CD3ε链、CD3δ链与CD3ε链均无相关性;铅中毒患者驱铅治疗前CD3γ链与CD3δ链,CD3γ链与CD3ε链,CD3δ链与CD3ε链均呈显著正相关,驱铅治疗后仅CD3δ链与CD3ε链呈显著正相关。结论铅中毒患者CD3γ、CD3δ、CD3ε链mRNA水平呈不同趋势的改变,提示铅中毒后对机体免疫系统有一定影响并且在不同基因之间存在差异;驱铅治疗后体内铅负荷降低,CD3δ链表达水平仍然与健康人不同,说明驱铅治疗后机体的免疫功能仍存在异常。
Objective To investigate the expression of CD3γ, CD3δ and CD3ε T-cell receptor signaling molecules in peripheral blood of lead poisoning patients after lead-removing treatment. Methods The mRNA levels of CD3γ, CD3δ and CD3ε in peripheral blood mononuclear cells were detected by real-time fluorescence quantitative PCR before and after lead-removing treatment in 9 healthy individuals and 16 patients with lead poisoning respectively. The β2 microglobulin (β2M) gene was Internal reference gene, according to the relative quantification formula 2- △ Ct × 100% calculate the gene expression levels. Spearman rank correlation was used to analyze the correlation between CD3γ, CD3δ and CD3ε mRNA levels in healthy people and lead poisoning patients before and after lead expelling treatment. Results There was no significant difference in CD3γ mRNA levels before and after lead poisoning in lead poisoning patients. The levels of CD3 δ mRNA in serum before and after lead removal were significantly lower than those in healthy controls. CD3 δ mRNA levels in lead poisoning patients were lower than those before treatment. CD3 epsilon chain mRNA level before extubation was significantly higher than that in healthy subjects, and CD3 epsilon chain mRNA level was restored after extubation of lead. There was no correlation between CD3γ chain and CD3δ chain, CD3γ chain and CD3ε chain, CD3δ chain and CD3ε chain in lead poisoning patients. CD3γ chain and CD3δ chain, CD3γ chain and CD3ε chain, CD3δ chain and CD3ε chain There was a significant positive correlation between CD3δ chain and CD3ε chain. Conclusions The levels of CD3γ, CD3δ and CD3ε chain mRNA in lead poisoning patients showed different trends, suggesting that lead poisoning may have some effects on the immune system and lead to differences in different genes.
