采用选择性抽提方法结合整装细胞电镜技术(whole mount cell EM)及扫描电镜技术,对单核、巨噬细胞的骨架系统进行了观察;并应用抗肌动蛋白(Anti-actin)抗体和抗α-管蛋白(Anti-αtubulin)单抗结合胶体金免疫电镜技术分别对其微丝、微管进行了定性研究。在此基础上,对不同剂量~(60)Coγ射线照射前后单核、巨噬细胞骨架系统的辐射效应进行了观察,发现照射后细胞骨架系统发生了显著的改变。同时,对照观察了秋水仙素、松胞素B对其骨架系统的影响,发现它们之间存在着一些类似之处。因此,我们认为~(60)Coγ射线引起的细胞骨架系统改变可能与微管、微丝的聚合障碍有关。
Using selective extraction methods combined with whole mount cell EM and scanning electron microscopy, the skeletal system of monocytes and macrophages was observed; anti-actin antibodies and The anti-α-tubulin monoclonal antibody and colloidal gold immunoelectron microscopy were used to qualitatively study the microfilaments and microtubules, respectively. On this basis, the radiation effects of mononuclear and macrophage cytoskeletal systems before and after irradiation with different doses of 60 Co γ-rays were observed, and it was found that the cytoskeletal system has undergone significant changes after irradiation. At the same time, the effects of colchicine and cytosine B on their skeletal system were observed and found to have some similarities. Therefore, we believe that the cytoskeletal system changes induced by ~(60)Co γ-rays may be related to the polymerization of microtubules and microfilaments.
