目的探讨接合物蛋白CRKL在肝癌中的表达及在肝癌细胞增殖中的作用。方法荧光定量PCR(real-time PCR)检测肝癌及癌旁组织中CRKL的mRNA的表达水平,同时检测肝癌细胞系及正常人肝细胞中CRKL的mRNA的表达水平,并通过在肝癌细胞株中过表达CRKL,分析CRKL蛋白对肝癌细胞增殖的作用。结果 48对样本中有31对CRKL的mRNA表达水平在肝癌组织中高于癌旁组织。在体外肝癌细胞的功能实验中,过表达CRKL能促进SK-hep-1和HepG2细胞的平板克隆及软琼脂克隆形成。结论 CRKL在肝癌组织中高表达,过表达CRKL能促进肿瘤的增殖,所以,CRKL可能成为临床治疗中抑制肿瘤生长的靶点。
Objective To investigate the expression of CRKL in hepatocellular carcinoma (HCC) and its role in the proliferation of hepatoma cells. Methods Real-time PCR was used to detect the mRNA expression of CRKL in hepatocellular carcinoma (HCC) and paracancerous tissues. Meanwhile, the mRNA expression of CRKL in HCC cell lines and normal human hepatocytes was detected. The expression of CRKL, analysis of CRKL protein on the proliferation of liver cancer cells. Results The mRNA expression of CRKL in 31 of 48 samples was higher than that in paracancerous tissues in HCC tissues. Overexpression of CRKL promoted platelet cloning and soft agar cloning of SK-hep-1 and HepG2 cells in vitro in functional assays of hepatoma cells. Conclusion CRKL is overexpressed in HCC tissues. Overexpression of CRKL can promote tumor proliferation. Therefore, CRKL may be the target of inhibiting tumor growth in clinical treatment.
