目的采用制备高效液相色谱法,从蒙药大栀子中分离纯化其主要化学成分环烯醚萜苷类和西红花苷类,建立其纯化工艺。方法大栀子醇提物采用柱色谱分离,然后采用制备色谱系统,收集对应峰的流出液,并对产品进行了纯度检测。经理化常数测定,结合现代波谱学技术确定其结构。结果经过分离鉴定,得到两大类化合物,即环烯醚萜苷类与西红花苷类,6个单体化合物:栀子苷、去乙酰车叶草苷酸甲酯、鸡矢藤次苷甲酯、京尼平-1-β-龙胆苷,西红花苷-1、西红花苷-2。除栀子苷外,其它5种化合物均为首次从大栀子中分离得到。结论利用制备色谱技术进行蒙药大栀子化学成分的分离和纯化,具有简便、快捷、周期短、自动化程度高、收集全面等优点。
OBJECTIVE To isolate and purify iridoid glycosides and crocin from the gardenia dahurica by preparative high performance liquid chromatography (HPLC). The purification process was established. Methods The crude extracts were separated by column chromatography, and then the chromatographic system was used to collect the effluent of the corresponding peaks. The purity of the products was tested. Managerial constant determination, combined with modern spectroscopy techniques to determine its structure. Results After isolation and identification, two major classes of compounds were obtained: iridoid glycosides and crocin, six monomer compounds: geniposide, deacetyl carotinyl glycoside methyl ester, Ester, genipin-1-β-gentinidin, crocin-1, crocin-2. In addition to geniposide, the other five compounds were isolated from the gardenia for the first time. Conclusion The separation and purification of the chemical constituents from the gardenia officinalis by preparative chromatography are simple, rapid, short cycle, high degree of automation, comprehensive collection and so on.
