目的:研究牡蛎钙补肾中药复方对骨形成蛋白-4(BMP-4)诱导成骨信号转导机制的调控作用。方法:采用切除大鼠双侧卵巢的方法复制绝经后骨质疏松症模型,应用牡蛎钙补肾中药复方进行防治,同时用骨疏康颗粒剂、牡蛎碳酸钙咀嚼片作为阳性对照,正常大鼠作为标准对照,模型大鼠作为模型空白对照。中药防治12周后进行取材及实验指标检测:用双能X线骨密度仪检测离体股骨骨密度;用W estern杂交法检测骨组织BMP-4和Sm ad5、6蛋白表达。结果:与模型空白组比较,牡蛎钙补肾中药复方各剂量组股骨骨密度显著升高(P<0.01);骨组织BMP-4、Sm ad5蛋白表达水平明显升高(P<0.01),Sm ad6蛋白表达水平明显降低(P<0.01)。结论:牡蛎钙补肾中药复方可明显上调BMP-4和Sm ad5蛋白表达,下调Sm ad6蛋白表达,这可能是其防治骨质疏松症的重要机制之一。
OBJECTIVE: To study the regulatory effects of oyster calcium and kidney-tonifying compound recipes on bone morphogenetic protein-4 (BMP-4)-induced osteogenesis signal transduction mechanism. METHODS: The model of postmenopausal osteoporosis was duplicated by removing the bilateral ovaries of the rats. The oyster calcium kidney tonic compound was used for prevention and treatment. Meanwhile, Gushukang granules and oyster calcium carbonate chewable tablets were used as positive controls. Normal rats were used as Standard controls, model rats served as model blank controls. After 12 weeks of Chinese medicine treatment, samples were taken and tested: bone density was measured by dual energy X-ray absorptiometry, and protein expression of BMP-4 and Sm ad5, 6 was detected by W estern hybridization. RESULTS: Compared with the model blank group, the BMD of the oyster calcium-reinforcing kidney Chinese medicine compound group significantly increased (P<0.01); the expression levels of BMP-4 and Sm ad5 protein in the bone tissue increased significantly (P<0.01). Protein expression was significantly reduced (P<0.01). Conclusion: Oyster calcium and kidney-tonifying Chinese herbal compound can significantly up-regulate the expression of BMP-4 and Sm ad5 protein, and down-regulate the expression of Sm ad6 protein, which may be one of the important mechanisms for prevention and treatment of osteoporosis.
