目的:探讨能全面地反映大黄中所含化学成分的指纹图谱的最适进样量。方法:采用HPLC法,系统的适应性条件:色谱柱为Alltech Apollo C18柱;流动相为乙腈-水-1%冰醋酸,梯度洗脱;检测波长为254nm;柱温为40℃。结果:从(22.41~276.24μg)(以大黄酸计算)分别进样,当仅以总信息熵作为参考时,其进样量为276.24μg时的信息熵最大,为3.293×1010,因此确定此条件下大黄指纹图谱的最佳进样量为276.24μg;但若以单位进样量的信息熵比较,那么以61.35μg的图谱所带的信息量最大,为3.354×109。因此确定此条件下大黄指纹图谱的最佳进样量为61.35μg。结论:信息熵可用来确定中药材指纹图谱的最佳进样量。
OBJECTIVE: To explore the optimum sample volume that can fully reflect the fingerprints of chemical constituents contained in rhubarb. Methods: The HPLC method was adopted. The system was adapted to the conditions: the column was Alltech Apollo C18 column; the mobile phase was acetonitrile-water-1% glacial acetic acid, gradient elution; the detection wavelength was 254 nm; the column temperature was 40°C. Results: From (22.41 to 276.24 μg) (calculated with Rhein), when the total information entropy was used as a reference, the information entropy when the sample volume was 276.24 μg was the highest, which was 3.293×1010. Therefore, confirm this. The optimal sample injection amount of rhubarb fingerprint is 276.24μg under the condition, but if the information entropy of unit injection amount is compared, then the information with 61.35μg map has the largest amount of information, which is 3.354×109. Therefore, the optimal injection volume for rhubarb fingerprinting under this condition was 61.35 μg. Conclusion: Information entropy can be used to determine the optimal injection volume of Chinese herbal medicine fingerprints.
