以2片真叶期的烟草单倍体幼苗为材料,采用二次饱和D-最优设计,研究了秋水仙素浓度及其浸苗处理时间对染色体加倍率及成苗的影响.结果表明:秋水仙素浓度、浸苗时间对加倍率、成苗数和双单倍体植株数的影响不一致;0.3469%的秋水仙素、浸苗72h处理的加倍率达到最高值54.18%;0.3878%的秋水仙素、浸苗24h的成苗数最多,达92.88株;0.3115%的秋水仙素、浸苗49.7h的双单倍体植株数最多,达37.06株.研究得出以获得最多的双单倍体系数量为研究目的时,采用0.3115%的秋水仙素、浸苗49.7h较为适宜.
Two true leaf tobacco haploid seedlings were used as materials to study the effect of colchicine concentration and its seedling treatment time on chromosome doubling rate and seedling formation by the second-order saturation D-optimal design.The results showed that: Colchicine concentration and soaking time had different effects on doubling rate, number of seedlings and number of double haploid plants. The doubling rate of 0.3469% colchicine and soaking seedlings reached the highest value 54.18% and 0.3878% Narcissus, the number of seedlings in soaking for 24 h was the highest, reaching 92.88; the number of double haploid plants with 0.3115% colchicine and soaking seedlings was 49.06 h, up to 37.06. The amount of system for the purpose of the study, the use of colchicine 0.3115%, Dip seedlings 49.7h more appropriate.
