目的:建立丹参白疕消丸同时测定盐酸小檗碱、丹皮酚、丹参酮ⅡA、大黄素、大黄酚和大黄素甲醚6种成分含量的HPLC测定方法。方法:采用高效液相色谱法,Inertsil ODS-3色谱柱(250 mm×4.6 mm,5μm),柱温25℃,流动相:甲醇-0.4%磷酸水溶液梯度洗脱,流速0.8 ml·min-1,检测波长为354 nm(盐酸小檗碱)、274 nm(丹皮酚)、254 nm(大黄酚、大黄素、大黄素甲醚和丹参酮ⅡA),外标法计算含量。结果:盐酸小檗碱、丹皮酚、大黄素、丹参酮ⅡA、大黄酚和大黄素甲醚6种成分分别在0.1939~1.5515,0.0417~0.3333,0.0625~0.5000,0.0784~0.6272,0.2120~1.6963,0.3584~1.0754μg与峰面积线性关系良好(r2>0.9982),样品平均回收率为95.67%~98.14%,RSD<1.85%。结论:该方法快速、简便、灵敏、重复性好,可用于丹参白疕消丸的质量控制方法。
OBJECTIVE: To establish an HPLC method for simultaneous determination of six components of berberine hydrochloride, paeonol, tanshinone Ⅱ A, emodin, chrysophanol and physcion in Salvia miltiorrhiza Baishaxiong pill. Methods: High performance liquid chromatography (HPLC) was performed on an Inertsil ODS-3 column (250 mm × 4.6 mm, 5 μm) with a column temperature of 25 ° C and a mobile phase of methanol-0.4% phosphoric acid , The detection wavelength was 354 nm (berberine hydrochloride), 274 nm (paeonol), 254 nm (chrysophanol, emodin, physcion and tanshinone Ⅱ A), the external standard method to calculate the content. Results: The contents of berberine, paeonol, emodin, tanshinone Ⅱ A, chrysophanol and physcion were 0.1939 ~ 1.5515,0.0417 ~ 0.3333,0.0625 ~ 0.5000,0.0784 ~ 0.6272,0.2120 ~ 1.6963,0.3584 ~ 1.0754μg had a good linear relationship with the peak area (r2> 0.9982). The average recoveries of samples were 95.67% ~ 98.14% with RSD <1.85%. Conclusion: The method is rapid, simple, sensitive and reproducible. It can be used for the quality control of Salvia Baishang Wan.
