目的利用响应面分析法对桦褐孔菌发酵培养基进行优化,提高多糖产量。方法应用Plackett-Burman设计和响应面法,对桦褐孔菌发酵培养基成分进行优化。用Design-Expert软件对实验数据进行多元回归分析,并建立3种主要因素(淀粉、酵母粉和磷酸二氢钠)与多糖产量之间的函数关系。用最终优化的配方进行3次验证实验。结果通过快速“登高法”对3个显著因素进行寻优,培养基中3因素最佳浓度为:淀粉7.5 g/L,酵母粉1.71 g/L,磷酸二氢钠1.35 g/L,胞外多糖产量最高可达9.12 g/L。3次验证实验所测得的多糖产量分别为9.08、8.96和9.12 g/L,平均值为9.05 g/L,与预测结果基本一致。结论优化的桦褐孔菌发酵培养基可显著提高胞外多糖产量。
Objective To optimize the fermentation broth of Inonotus obliquus using response surface methodology to improve the yield of polysaccharides. Methods Plackett-Burman design and response surface methodology were used to optimize the fermentation broth composition of Inonotus obliquus. Multiple regression analysis of the experimental data was performed using Design-Expert software and a functional relationship between the three main factors (starch, yeast powder and sodium dihydrogen phosphate) and polysaccharide yield was established. Three validation experiments with the final optimized formula. Results Three factors were optimized by fast “ascension” method. The optimal concentration of three factors in culture medium was 7.5 g / L starch, 1.71 g / L yeast, 1.35 g / L sodium dihydrogen phosphate, Extracellular polysaccharide production up to 9.12 g / L. The yield of polysaccharides measured by the three validation experiments were 9.08, 8.96 and 9.12 g / L, respectively, with an average value of 9.05 g / L, which was consistent with the predicted results. Conclusion The optimized fermentation broth of Inonotus obliquus can significantly improve the yield of extracellular polysaccharide.
