高效免疫抑制剂环孢霉素A(cyclosporineA,CsA)和FK506主要是通过阻滞IL-2基因表达而实现其免疫抑制作用。至于它们是作用于细胞因子基因的转录,还是基因转录的信号传导途径,一直是细胞生物学家和免疫学家关注的课题。近年来由于分子生物学和免疫学的发展,这方面的研究已取得了迅速的进展,本文着重介绍CsA和FK506的胞浆受体亲环肽素和FK506结合蛋白的生化特性、生物学功能以及这种免疫抑制剂对Ca~(++)相关信号传导途径的影响来说明其可能的免疫抑制作用机制。
Efficient immunosuppressants cyclosporine A (CsA) and FK506 mainly achieve their immunosuppressive effects by blocking the expression of IL-2 gene. Whether they act on the transcription of cytokine genes or on the signaling pathways of gene transcription has long been a concern for cell biologists and immunologists. In recent years, due to the development of molecular biology and immunology, rapid progress has been made in this aspect. This article focuses on the biochemical characteristics, biological functions and the biological activities of the cyclosporin and FK506 binding proteins of CsA and FK506 The effect of this immunosuppressant on the Ca ~ (++) - related signaling pathway indicates its possible immunosuppressive mechanism.
