目的:评价不同生长期及不同年限葛根多糖含量的动态积累。方法:以葡萄糖为对照品,采用以苯酚-硫酸法,在486 nm处测定不同生长期及不同年限葛根中多糖的含量。结果:在0.020~0.142 mg.L-1葡萄糖质量浓度与吸光度线性关系良好(r=0.999 9),平均回收率99.34%,RSD 1.82%。葛根中多糖的含量呈规律性变化,每年以8,9月份葛根多糖的含量较高,分别为10.28%和11.35%。多年生葛根中多糖含量明显高于前3年,为14.28%。结论:该文结果可为野葛规范化种植及药材采收加工提供科学依据。
Objective: To evaluate the dynamic accumulation of polysaccharide in Radix Puerariae at different growth stages and different years. Methods: Glucose was used as reference substance, and the content of polysaccharide in Radix Puerariae was determined by phenol-sulfuric acid method at 486 nm for different growth periods and different years. Results: There was a good linear relationship between absorbance and concentration of 0.020-0.142 mg.L-1 glucose (r = 0.999 9). The average recovery was 99.34% and the RSD was 1.82%. The contents of polysaccharides in Pueraria lobata showed regular changes. The content of Pueraria lobata polysaccharides in August and September each year was 10.28% and 11.35% respectively. Perennial Pueraria polysaccharide content was significantly higher than the previous 3 years, 14.28%. Conclusion: The results of this paper can provide a scientific basis for standardized cultivation of kudzu and harvesting and processing of medicinal materials.
