目的:观察Gq/11蛋白、心功能在急性呼吸窘迫综合征(acuterespiratorydistresssyndrome,ARDS)过程所继发的心脏损害中的变化及异丙酚预处理后的变化,探讨ARDS过程Gq/11蛋白在心功能障碍中的作用机制及异丙酚对心功能的保护效应。方法:采用尾静脉注射油酸(oleicacid,OA)法复制大鼠ARDS模型,将24只健康雄性Wistar大鼠随机分为对照组(C组)、实验组(OA组)和异丙酚预处理组(P组)。所有大鼠经右颈动脉置管检测心功能指标,同时检测血浆及心肌组织乳酸脱氢酶(LDH)、肌酸磷酸激酶(CK)、丙二醛(MDA)含量以及免疫印迹法测定心肌组织Gq/11蛋白含量。结果:左室内压(LVSP)在OA组、P组均下降,与C组相比差异具有统计学意义(P<0.01);左室最大上升速率(+dp/dtmax)在OA组下降,与C组相比差异具有统计学意义(P<0.01)。左室最大下降速率(dp/dtmax)呈现与LVSP相同的变化。血浆LDH水平在OA组升高,与C组、P组相比差异具有统计学意义(P<0.01);心肌组织LDH水平在C组最低,在OA组最高,P组居中,实验各组之间差异具有统计学意义(P<0.01)。血浆及心肌组织CK、MDA水平呈现与心肌组织LDH相同的变化;Gq/11蛋白含量在OA组、P组均升高,与C组相比差异具有统计学意义(P<0.01)。结论:Gq/11蛋白的表达上调在ARDS过程继发的心脏损害中起关键作用,预?
OBJECTIVE: To observe the changes of Gq / 11 protein and cardiac function in heart damage secondary to acute respiratory distress syndrome (ARDS) and the changes after propofol preconditioning, and to explore the role of Gq / 11 protein in cardiac function The mechanism of action and the protective effect of propofol on cardiac function. Methods: The rat model of ARDS was established by oleic acid (OA) injection. 24 healthy male Wistar rats were randomly divided into control group (C group), experimental group (OA group) and propofol pretreatment Group (Group P). All the rats were tested for cardiac function by right carotid artery catheterization, and the content of lactate dehydrogenase (LDH), creatine phosphokinase (CK) and malondialdehyde (MDA) in plasma and myocardium were measured simultaneously, and the myocardial tissue Gq / 11 protein content. Results: The left ventricular pressure (LVSP) decreased in both OA and P groups, with statistical significance (P <0.01).
