为提高桑黄液体菌丝体中粗多糖产量,利用SAS软件对桑黄菌液体发酵培养条件进行优化研究。运用Plackett-Bunnan设计法筛选出影响桑黄粗多糖产量的主要因素,在此基础之上应用中心组合进行3因素3水平的试验设计,以桑黄胞外多糖产量为响应值,应用响应面法(RSM)对桑黄液体发酵培养条件进行进一步的优化。结果表明,当装液量127 mL/500 mL、pH5.45、摇床转速175 r/min时,桑黄胞外粗多糖产量达到最大值6.27 g/L,较优化前的5.14 g/L提高了22%。
In order to improve the crude polysaccharides yield in the mycelium of Phellinus igniarius, the fermentation conditions of Phellinus igniarius liquid were optimized by SAS software. Plackett-Bunnan design was used to screen out the main factors that affect the yield of polysaccharides from Phellinus igniarius. Based on this, three-factor-three experimental design was applied to the center combinations. The yield of polysaccharide was determined by response surface methodology (RSM) to further optimize the fermentation conditions of Phellinus igniarius liquid. The results showed that when the liquid volume was 127 mL / 500 mL, pH5.45 and the shaking speed was 175 r / min, the yield of polysaccharides from Pseudomonas solanacearum reached 6.27 g / L, which was 5.14 g / L before optimization 22%.
