目的:建立高效液相色谱法同时测定矮林杜仲叶中6种成分的含量。方法:采用Thermo BDS HYPERSIL C18色谱柱(250 mm×4.6 mm,5μm),以甲醇(A)-0.05%磷酸(B)为流动相,梯度洗脱,程序化变波长,体积流量0.8 m L/min,柱温35℃。结果:桃叶珊瑚苷、京尼平苷酸、绿原酸、京尼平苷、松脂醇二葡萄糖苷、芦丁6种成分的质量浓度分别在1.0~500.0μg/m L、5.0~600.0μg/m L、4.0~300.0μg/m L、0.2~50.0μg/m L、0.2~150.0μg/m L、0.1~100.0μg/m L内与峰面积呈良好的线性关系,平均加样回收率分别为102.3%、99.9%、100.8%、99.8%、100.6%、99.1%,其RSD分别为4.4%、0.6%、4.1%、1.0%、0.7%、5.0%。结果表明,不同采收期矮林杜仲叶中桃叶珊瑚苷、京尼平苷酸、绿原酸等主要有效成分的含量有明显差别。结论:该方法重复性好,可操作性强,可用于矮林杜仲叶中6种有效成分的含量测定,可较全面地反映不同采收时期矮林杜仲叶中有效成分的差异,通过对结果分析,8月底采收的矮林杜仲叶的整体质量最好。
Objective: To establish a method for the simultaneous determination of six components in Eucommia ulmoides leaves by HPLC. Methods: The mobile phase was eluted with a gradient of methanol (A) - 0.05% phosphoric acid (B) using a Thermo BDS HYPERSIL C18 column (250 mm × 4.6 mm, 5 μm) min, column temperature 35 ℃. Results: The concentrations of aucubin, genipin, chlorogenic acid, geniposid, pinorelin diglucoside and rutin were 1.0-500.0μg / mL, 5.0-600.0μg There was a good linear relationship between the concentration and peak area in the range of 4.0-300.0μg / mL, 0.2-50.0μg / mL, 0.2-150.0μg / mL and 0.1-100.0μg / mL. The average recovery was The RSDs were 102.3%, 99.9%, 100.8%, 99.8%, 100.6% and 99.1%, respectively, with RSDs of 4.4%, 0.6%, 4.1%, 1.0%, 0.7% and 5.0% respectively. The results showed that the contents of aucubin, genipin acid, chlorogenic acid and other major active ingredients in the leaves of Eucommia ulmoides differed significantly in different harvest time. Conclusion: The method is reproducible and operable, and can be used to determine the content of six active ingredients in Eucommia ulmoides leaves, which can reflect the difference of active ingredients in Euonymus japonicus leaves in different harvest time more completely.
