目的观察埃兹蛋白(ezrin)在前列腺癌组织中的表达,探讨ezrin基因沉默对人前列腺癌PC-3细胞增殖、侵袭的影响及相关机制。方法收集前列腺癌和良性前列腺增生患者的标本各20例,采用荧光实时定量PCR检测ezrin mRNA水平、Western blot法检测ezrin蛋白水平。培养PC-3细胞,分别用培养液、Scramble siRNA、ezrin siRNA处理48 h;MTT法检测细胞增殖情况,TranswellTM侵袭实验检测细胞侵袭能力,荧光实时定量PCR、Western blot法检测上皮钙黏素、神经钙黏素的mRNA和蛋白水平。结果与良性前列腺增生患者的组织相比,前列腺癌组织中ezrin mRNA、蛋白表达水平均明显增加。与Scramble siRNA处理的PC-3细胞相比,ezrin表达下调后,PC-3细胞增殖受到明显抑制,平均穿膜细胞数、神经钙黏素水平降低,上皮钙黏素水平增加。Scramble siRNA处理的PC-3细胞与单纯培养液处理的PC-3细胞上述指标无明显差异。结论 Ezrin在前列腺癌组织中高表达,ezrin基因沉默能抑制PC-3细胞的增殖和侵袭,同时上调上皮钙黏素表达及下调神经钙黏素表达。
Objective To observe the expression of ezrin in prostate cancer and to explore the effect and mechanism of ezrin gene silencing on the proliferation and invasion of human prostate cancer PC-3 cells. Methods Twenty samples of prostate cancer and benign prostatic hyperplasia were collected. The ezrin mRNA level was detected by real-time fluorescence quantitative PCR. The ezrin protein level was detected by Western blot. The PC-3 cells were cultured for 48 h with culture medium, Scramble siRNA and ezrin siRNA respectively. The proliferation of PC-3 cells was detected by MTT assay. The cell invasion was detected by TranswellTM invasion assay. The expressions of E-cadherin, Cadherins mRNA and protein levels. Results Compared with the tissues of benign prostatic hyperplasia patients, the ezrin mRNA and protein expression levels in prostate cancer tissues were significantly increased. Compared with PC-3 cells treated with Scramble siRNA, the proliferation of PC-3 cells was significantly inhibited after ezrin expression was down-regulated. The average number of transmembrane cells, neuronal cadherin, and cadherin increased. Scramble siRNA-treated PC-3 cells and PC-3 cells treated with simple culture medium no significant difference in the above indicators. Conclusion Ezrin is overexpressed in prostate cancer tissues. Ezrin silencing can inhibit the proliferation and invasion of PC-3 cells, upregulate the expression of E-cadherin and down-regulate the expression of E-cadherin.
