目的:研究人热休克肿瘤抗原复合物对树突状细胞(dendritic cells,DC)表面共刺激分子表达的影响。方法:分离培养人外周血来源的DC细胞,在培养过程中加入分离纯化的人热休克蛋白70(HSP70)抗原复合物及肿瘤细胞裂解物,流式细胞技术分析比较负载HSP70肿瘤抗原复合物、负载肿瘤细胞裂解物及负载前DC细胞表面分子CD80、CD83、CD86及HLA-DR表达水平的变化。结果:培养基中加入HSP70肿瘤抗原复合物后,其表面共刺激分子CD80、CD86和成熟标记分子CD83的表达水平分别为(98.9±1.5)%、(98.3±1.3)%和(53.1±0.8)%,与负载前CD80、CD86和CD83的(66.2±1.6)%、(94.2±2.0)%和(16.3±1.4)%相比显著提高,P=0.013;与肿瘤细胞裂解物负载的DC表面CD80、CD86和CD83的(86.2±1.9)%、(96.5±1.1)%和(55.1±1.0)%相比,共刺激分子CD80的表达显著提高,P=0.020。结论:人热休克肿瘤抗原复合物可提高DC表面共刺激分子的表达水平,促进DC功能活化,有可能在抗肿瘤免疫机制中起重要作用。
Objective: To study the effect of human heat shock tumor antigen complex on costimulatory molecule expression on dendritic cells (DCs). Methods: DCs isolated from human peripheral blood were isolated and cultured. Purified human HSP70 antigen complex and tumor cell lysate were added into the culture process. Flow cytometry was used to analyze the expression of HSP70 tumor antigen complex, The changes of the expression of CD80, CD83, CD86 and HLA-DR on the surface of tumor cells loaded with lysate and pre-loaded DCs. Results: The expression of costimulatory molecules CD80, CD86 and mature marker CD83 were (98.9 ± 1.5)%, (98.3 ± 1.3)% and (53.1 ± 0.8)%, respectively, %, Significantly increased compared to (66.2 ± 1.6)%, (94.2 ± 2.0)% and (16.3 ± 1.4)%, respectively, of pre-loaded CD80, CD86 and CD83 with P = 0.013; CD80 with DCs loaded with tumor cell lysate , CD86 and CD83 (86.2 ± 1.9)%, (96.5 ± 1.1)% and (55.1 ± 1.0)%, the co-stimulatory molecule CD80 expression was significantly increased, P = 0.020. Conclusion: Human heat shock tumor antigen complex can increase the expression of costimulatory molecules on DCs and promote the activation of DCs, which may play an important role in anti-tumor immune mechanisms.
