目的 提取牛骨形态发生蛋白 ,并观察其对人骨髓基质细胞的体外成骨分化作用 ,为将骨形态发生蛋白与骨髓基质细胞用于进一步的骨缺损修复研究提供实验依据。方法 从小牛骨中提取骨形态发生蛋白 ,配制条件培养液 ,并作用于培养状态的人骨髓基质细胞 ,染色检测细胞碱性磷酸酶、Ⅰ型胶原表达情况 ,放免法测定培养液中骨钙素含量。结果 人骨髓基质细胞经诱导培养后 ,碱性磷酸酶、Ⅰ型胶原、骨钙素表达明显增加。结论 体外培养时 ,天然骨形态发生蛋白可促进人骨髓基质细胞的成骨分化。
OBJECTIVE: To extract the bovine bone morphogenetic protein (BMP) and to observe the osteogenic differentiation of human bone marrow stromal cells in vitro, so as to provide experimental evidence for the study of bone morphogenetic protein and bone marrow stromal cells for further study of bone defect repair. Methods Bone morphogenetic protein was extracted from calf bones and conditioned medium was prepared and cultured in human bone marrow stromal cells. The expression of alkaline phosphatase and type Ⅰ collagen was detected by staining, and the osteocalcin content. Results After induction of human bone marrow stromal cells, the expression of alkaline phosphatase, type Ⅰ collagen and osteocalcin increased significantly. Conclusion In vitro culture, natural bone morphogenetic protein can promote the osteogenic differentiation of human bone marrow stromal cells.
