目的探讨用HCMVAD169病毒株建立小鼠感染动物模型的可行性。方法 BALB/c近交系小鼠30只,随机分为三组,分别为病毒感染组、免疫抑制病毒感染组、正常对照组,病毒接种途径为腹腔注射。观察动物的变化,取各组小鼠全血分离有核细胞,用直接荧光免疫法检测CMVpp65早期抗原,以病理学方法检测动物组织器官的损伤特点。结果接种了病毒的二组小鼠都出现了明显体重不增、皮毛稀松、生长迟缓等病症,肺、肠、涎腺等组织器官发生了广泛病理损害,免疫抑制病毒感染组小鼠血液有核细胞内发现了pp65抗原阳性细胞。结论将HCMVAD169株接种BALB/c近交系小鼠建立感染模型是可行的,在机体免疫抑制状态HCMV可激活感染。
Objective To investigate the feasibility of using HCMVAD169 strain to establish a mouse model of infection in mice. Methods Thirty BALB / c inbred mice were randomly divided into three groups: viral infection group, immunosuppressive virus infection group and normal control group. The route of virus inoculation was intraperitoneal injection. The changes of animals were observed. The whole blood of each group was used to separate the nucleated cells. The direct anti-CMVpp65 antigen was detected by direct immunofluorescence assay. The damage characteristics of tissues and organs were detected by pathology. Results The two groups of mice inoculated with virus showed obvious weight loss, loose slippage and growth retardation, extensive pathological damage in tissues and organs such as lung, intestine and salivary gland, blood in mice with immunosuppressive virus infection Pp65 antigen-positive cells were found in the cells. Conclusion Inoculation of HCMVAD169 strain in BALB / c inbred mice is feasible to establish the infection model. HCMV can activate infection in immunosuppressed state.
