目的 通过流式原位杂交法 (FLOW FISH)研究白血病和骨髓异常增生综合征 (MDS)患者骨髓中白细胞端粒长度的变化。方法 采用荧光原位杂交法结合流式细胞术检测 4 3例血液疾病患者和 4 8名健康对照者骨髓中白细胞的端粒长度。以流式细胞仪测定的细胞平均荧光强度 ,即Q FISH值来表示端粒长度。结果 健康对照组Q FISH值为 0 0 66± 0 0 15 ,与同年龄健康对照相比较 ,除 3例未发生转型的MDS和 2例缓解期慢性髓细胞性白血病 (CML)患者的端粒长度无明显变化外(Q FISH值分别为 0 0 69± 0 0 0 7和 0 0 60± 0 0 0 8) ,其余 3 8例患者 (包括 2例转型为急性髓细胞性白血病的MDS和 2例急变期CML)骨髓中白血病细胞的端粒长度均明显变短 (P <0 0 5 ) ,Q FISH值为0 0 4 7± 0 0 19。 8例白血病患者在初诊和化疗完全缓解后端粒长度的改变无统计学差异 (P >0 0 5 ) ,但慢性淋巴细胞性白血病 (CLL)治疗后端粒有缩短趋势 (由 0 0 5 2变为 0 0 4 8) ,而急性髓细胞性白血病 (AML)治疗后端粒有变长趋势 (由 0 0 4 3变为 0 0 5 6)。健康对照组端粒长度随年龄增加而变短 (r=0 67) ,而患病组端粒长度与年龄不相关 (r =0 14 )。结论 通过FLOW FISH法检测到大部分白血病和MDS患者骨髓中端粒长度比健康人短 ,其?
Objective To investigate the leukocyte telomere length changes in bone marrow of patients with leukemia and myelodysplastic syndrome (MDS) by flow in situ hybridization (FLOW FISH). Methods The telomere length of leukocytes in 43 patients with blood diseases and 48 healthy controls were detected by fluorescence in situ hybridization and flow cytometry. The telomere length is expressed as the mean fluorescence intensity measured by flow cytometry, ie Q FISH value. Results The QFISH value of healthy control group was 0 66 ± 0 0 15. Compared with healthy controls of the same age, the telomere length of 3 patients without MDS and 2 patients with remittent chronic myelogenous leukemia (CML) The QFISH values were 0 0 69 ± 0 0 0 7 and 0 0 60 ± 0 0 0 8, respectively. The telomere length of healthy control group was shorter with age (r = 0 67), while the telomere length in diseased group was not related to age (r = 0 14).
