目的观察成年鼠糖尿病康自发复时胰岛β细胞特异基因的表达。方法建立自发康复糖尿病成年小鼠模型。在糖尿病自发康复过程中,以流式细胞术分选其胰岛β细胞,通过mRNA差异显示法研究胰岛β细胞基因差异表达,以免疫荧光双标显示其蛋白差异表达。结果在糖尿病自发康复过程中,以mRNA差异显示法在胰岛β细胞内共检出12个差异表达基因,其中,雄激素受体(AR)在糖尿病最严重时低表达甚至不表达,在糖尿病康复过程中表达增强,在糖尿病完全康复时高表达(P<0.01)。结论以mRNA差异显示法研究胰岛β细胞特异基因的表达切实可行。糖尿病康复时,胰岛β细胞雄激素受体表达与功能性胰岛β细胞的增多密切相关。
Objective To observe the expression of islet β cell-specific genes in spontaneous rehospitalization of diabetic rats. Methods A spontaneous diabetic model of adult mice was established. During the spontaneous recovery of diabetes mellitus, the islet β cells were sorted by flow cytometry. The differentially expressed genes of pancreatic β cells were studied by mRNA differential display. The protein was differentially expressed by double immunofluorescence staining. Results During the spontaneous recovery of diabetes mellitus, 12 differentially expressed genes were detected in pancreatic β cells by mRNA differential display. Among them, androgen receptor (AR) was low or not expressed in the most severe stage of diabetes. The expression was enhanced in the course of diabetes mellitus and was highly expressed in the complete recovery of diabetes (P <0.01). Conclusion It is practicable to study the expression of islet β cell specific genes by mRNA differential display. During the recovery of diabetes, the expression of androgen receptor in pancreatic β cells is closely related to the increase of functional islet β cells.
