目的:观察伯氏疟原虫(P.berghei)小亚单位核糖体RNA(SSUrRNA)在斯氏按蚊体内的转录类型和动态。方法:应用RT-PCR方法,特异地扩增伯氏疟原虫A型和S型SSUrRNA,检测斯氏按蚊虫体内S型SSUrRNA的转录水平。结果:吸感染血的斯氏按蚊体内仅见S型SSUrRNA的条带。整体检测从第2天开始查见,第8天明显增强,第12~16天达到高峰。在蚊腹部S型SSUrRNA转录动态与整体检测结果相似。胸部于第12天查见该基因的转录。结论:斯氏按蚊体内伯氏疟原虫仅转录S型SSUrRNA,它的转录动态与疟原虫在蚊体内的发育密切相关。
Objective: To observe the transcriptional types and dynamics of P. berghei small subunit ribosomal RNA (SSUrRNA) in An. Methods: RT-PCR was used to amplify the type A and type S SSR rRNA of Plasmodium berghei to detect the transcription level of S-type SSUrRNA in the mosquitoes. Results: Only S-type SSUrRNA bands were found in the infected Anopheles stephensi. The overall test from the first two days of inspection, significantly enhanced on the 8th, the first 12 to 16 days to reach the peak. The transcriptional activity of S-type SSUrRNA in mosquito abdomen was similar to the overall test. The thorax was found on the 12th day of the gene transcription. Conclusion: Plasmodium berghei in S. anopheles only transcribe S-type SSUrRNA, and its transcriptional activity is closely related to the development of Plasmodium in the mosquito.
