目的观察脂肪细胞经不同浓度葡萄糖和胰岛素干预后内脂素表达的改变,探讨内脂素在糖尿病发病过程中的作用。方法培养3T3-L1前脂肪细胞,并诱导分化成成熟脂肪细胞。采用不同浓度葡萄糖、胰岛素孵育脂肪细胞48 h,RT-PCR和western blot技术检测3T3-L1脂肪细胞中内脂素mRNA和蛋白的表达水平。结果与对照组比较,随着葡萄糖浓度的增加,3T3-L1脂肪细胞内脂素mRNA和蛋白的表达量下降(P均<0.05);在不同浓度胰岛素作用下,3T3-L1脂肪细胞内脂素mRNA和蛋白的表达量改变无统计学意义(P>0.05)。结论高糖状态可抑制体外培养的脂肪细胞内脂素的表达,胰岛素浓度的改变对内脂素表达无影响。
Objective To observe the changes of visfatin expression in adipocytes treated with different concentrations of glucose and insulin to explore the role of visfatin in the pathogenesis of diabetes mellitus. Methods 3T3-L1 preadipocytes were cultured and differentiated into mature adipocytes. Adipocytes were incubated with different concentrations of glucose and insulin for 48 h. The expression of visfatin mRNA and protein in 3T3-L1 adipocytes was detected by RT-PCR and western blot. Results Compared with the control group, the expression of visfatin mRNA and protein in 3T3-L1 adipocytes decreased with the increase of glucose concentration (P <0.05). Under the action of different concentrations of insulin, There was no significant difference in mRNA and protein expression (P> 0.05). Conclusion High glucose can inhibit the expression of visfatin in adipocytes cultured in vitro. The change of insulin concentration has no effect on visfatin expression.
