目的 检测肝癌中 Ecadherin 的突变情况,探讨 Ecadherin 与肝癌发生和发展的关系。方法 采用 P C R S S C P方法,分析了 12 例肝癌旁组织、5 例肝癌栓组织和 22 例肝癌组织 D N A 样品中 Ecadherin 基因第 6、9 和 10 外显子 D N A 单链构象多态性。结果 12 例肝癌旁组织和 4 例分化较好的肝癌组织中, Ecadherin 基因第 6、9 和 10 外显子均未见有异常。在 18 例恶性程度较高的肝癌组织中发现 4 例突变,其中 2 例突变发生在第 9 外显子,2 例突变发生在第 6外显子。5 例肝癌栓组织中检出 2 例突变(占 40% ),其中 1例突变在第 9 外显子,另1 例突变发生在第 10 外显子。结论 Ecadherin 基因随肝癌的恶化,特别随转移潜能的获得,而发生突变频率的增加,表明它可能与肝癌的发展和转移相关。
Objective To detect the mutation of Ecadherin in HCC and explore the relationship between Ecadherin and the occurrence and development of HCC. METHODS: P C R S S C P method was used to analyze exon 6, 9 and 10 exon D of E cadherin gene in 12 cases of hepatocellular carcinoma, 5 cases of hepatocellular carcinoma and 22 cases of hepatocellular carcinoma. N A single strand conformation polymorphism. Results There were no abnormalities in the 6th, 9th and 10th exons of Ecadherin gene in 12 cases of hepatocellular carcinoma and 4 cases of well differentiated hepatocellular carcinoma. Four mutations were found in 18 cases of hepatocellular carcinoma with high degree of malignancy. Two of the mutations occurred in the 9th exon, and the other 2 mutations occurred in the 6th exon. Two cases of mutations (40%) were detected in 5 cases of hepatocellular carcinoma. One of the mutations was in the 9th exon, and the other mutation was in the 10th exon. Conclusion The E-cadherin gene is associated with the deterioration of liver cancer, especially with the acquisition of metastatic potential, and the frequency of mutations increases, indicating that it may be related to the development and metastasis of liver cancer.
