目的 建立一种简便、准确、快速检测微量血红蛋白 (HGB)的检验方法。方法 用淋巴细胞杂交瘤技术制备的特异性抗人HGB单克隆抗体 ,在ELISA双抗体夹心的基础上建立快速一步法 ,用于检测粪便中微量HGB抗原。结果 检测灵敏度可达 0 .0 5 8μg/ml,批内及批间变异系数分别为 4.2 %和 5 .1% ;并与常规ELISA法进行了相关分析 ,相关系数r =0 .96。经临床应用 ,效果良好。结论 该方法可作为早期快速诊断胃肠道出血性疾病的重要方法。
Objective To establish a simple, accurate and rapid test method for the detection of trace hemoglobin (HGB). METHODS: The anti-human HGB monoclonal antibody prepared by lymphocyte hybridoma technique was used to establish a rapid one-step method based on ELISA double antibody sandwich to detect trace HGB antigen in feces. Results The detection sensitivity was 0. 0 5 8μg/ml, intra-assay and inter-assay coefficients of variation were 4.2% and 5.1%, respectively, and correlation analysis was performed with conventional ELISA. The correlation coefficient was r = 0.96. After clinical application, the effect is good. Conclusion This method can be used as an early method for rapid diagnosis of gastrointestinal bleeding.
