本文分别用转移印迹杂交和斑点杂交检测61例乙肝患者外周血单个核细胞(PBMC)及血清中的HBV-DNA。同时用转移印迹杂交和原位杂交检测其中31例患者肝活检组织中的HBV-DNA。结果发现血清、PBMC及肝组织HBV-DNA阳性率分别为26.2%(16/61),24.6%(15/61)及48.4%(15/31)。同一个体内,血清、PBMC及肝组织内HBV-DNA之间无明确的关联,血清HBV-DNA阴性时,仍可在11例患者的PBMC或肝内检出HBV-DNA。结果表明,PBMC可能也是独立于肝外的HBV贮存及复制的场所。血清HBV-DNA阴性不能否定体内仍有HBV复制的可能。
In this paper, 61 cases of hepatitis B patients with peripheral blood mononuclear cells (PBMC) and serum HBV-DNA were detected by transfer blot hybridization and dot blot hybridization. At the same time, HBV-DNA in liver biopsies of 31 patients was detected by transfer blot hybridization and in situ hybridization. The positive rates of HBV DNA in serum, PBMC and liver tissues were 26.2% (16/61), 24.6% (15/61) and 48.4% (15/31) respectively. There was no clear correlation between HBV-DNA in serum, PBMC and liver tissue in the same individual. HBV-DNA could still be detected in PBMC or liver of 11 patients when serum HBV-DNA was negative. The results show that PBMC may also be independent of extrahepatic HBV storage and replication of the place. Serum HBV-DNA negative can not deny the possibility of HBV replication in vivo.
