环磷酰胺对不同等级SD大鼠胚胎骨骼发育的影响

目的观察环磷酰胺对SPF级、普通级SD大鼠胚胎骨骼发育的影响。方法采用SD大鼠致畸实验方法。分组:SPF级孕鼠22只为A组(A1、A2组,每组11只),普通级孕鼠20只为B组(B1、B2组,每组10只)。A1、B1组于孕第13天腹腔注射生理盐水1ml/100g bw 1次,A2、B2组于孕第13天腹腔注射环磷酰胺12.5mg/kg bw 1次。各组孕鼠第18天麻醉、解剖、取出胚胎,测量胚胎的基本发育指标,通过骨骼双染法对部分胚胎进行各部骨发育情况观察。结果 SPF级、普通级两大组在胚胎整体发育中除胚胎体重在2个NS组间有差异(P<0.01)外,其余各项指标仅在两大组内的NS组与CP组间之间有显著性差异(P<0.01)。在胚胎骨骼发育中胸骨节骨化点仅在SPF级组内的NS、CP组间之间有差异(P<0.01)外,枕骨评分、胸骨节数、尾椎数及全部四肢长骨骨化长度均在SPF级、普通级两大组内的NS组与CP组间之间均有显著性差异(P<0.01),而所有组别的肋骨数、颈椎数、胸椎数、腰椎数、荐骨数均无差异(P>0.05)。结论本实验条件下NS组胚胎的胎盘重量与胸骨骨化点数在SPF级比普通级发育的要早一点,其它指标及环磷酰胺诱导下的胚胎发育情况在两大组(A、B)之间基本一致。建议致畸实验尽可能选用SPF级SD大鼠。 Objective To observe the effects of cyclophosphamide on embryonic skeletal development of SPF-class and general-grade SD rats. Methods The method of teratogenicity in SD rats was used. Groups: 22 pregnant rats of group SPF were divided into group A (group A1 and group A2, 11 rats in each group), and group B was divided into group B (group B1 and group B2, 10 rats in each group). Groups A1 and B1 were intraperitoneally injected with 1 ml / 100 g bw of physiological saline on the 13th day of pregnancy, and groups A2 and B2 were intraperitoneally injected with 12.5 mg / kg bw of cyclophosphamide on the 13th day of pregnancy. The pregnant rats in each group were anesthetized and dissected on the 18th day, the embryos were removed, the basic developmental indexes of the embryos were measured, and the bone development of each part of the embryos was observed by the bone double staining method. Results There was significant difference between the two groups (SPF level and common level) except embryo body weight between the two NS groups (P <0.01) There was a significant difference (P <0.01). In the development of embryonic skeletal sternum only in the SPF group within the NS and CP groups between the differences (P <0.01), the occipital score, the number of sternum, the number of caudal vertebrae and all the limb long bone ossification length There was significant difference between NS group and CP group in SPF level and general level (P <0.01), while the number of ribs, cervical vertebra, thoracic vertebra, lumbar vertebra, sacral bone There was no difference in number (P> 0.05). CONCLUSIONS: The placental weights and sarcopenia scores of NS group embryos were slightly earlier than those of common stage under the experimental conditions. Other indicators and embryonic development induced by cyclophosphamide were significantly different between the two groups (A, B) Between basically the same. Proposed teratogenic test as possible selection SPF SD rats.
