对从宁夏平罗、中宁两个地区收集的当年生沙枣种子,分别用含有不同浓度Na Cl(0、0.3%、0.6%、0.9%和1.2%,m/V)的1/5(V/V)Hoaglan d溶液处理,以探讨这两个地区沙枣种子在萌发阶段对盐胁迫的响应特性及可能原因。结果表明,随着Na Cl浓度的升高,中宁和平罗沙枣种子的萌发率均逐渐下降,与中宁的沙枣种子相比,平罗的沙枣种子表现出更强的抗盐能力。伊文斯蓝(Evans blue)染色和质膜透性实验结果表明,随着Na Cl浓度的增加,中宁和平罗沙枣的种子胚的细胞膜透性逐渐增加,即Na Cl胁迫破坏了种子胚的质膜选择透性,使其增大,细胞内大量电解质外渗,破坏了细胞的稳态。通过Li Cl溶液和等渗的甘露醇溶液处理两个地区沙枣种子并测定其萌发率表明,Na Cl对沙枣种子萌发的抑制既有渗透效应又有离子效应,但是以渗透效应为主;平罗的沙枣种子表现出更强的耐盐能力,是因为它有更高的抗渗透胁迫能力。
Seeds collected from two areas of Pingluo and Zhongning in Ningxia were treated with different concentrations of NaCl (0, 0.3, 0.6, 0.9 and 1.2%, m / V) V / V) Hoaglan d solution to investigate the response characteristics and possible causes of the jujube seeds in the two germination stages. Evans blue staining and plasma membrane permeability test results showed that with the increase of NaCl concentration, the membrane permeability of seed embryos of Zhongning and Luozao jujube increased gradually, that is, NaCl stress destroyed the seed embryos Plasma membrane permeability, increase it, a large number of electrolyte extravasation within the cell, undermining the cell’s homeostasis. The treatment of jujube seeds in two regions by Li Cl solution and isotonic mannitol solution and the determination of their germination rate showed that Na Cl inhibited osmotic and ionogenic activities of seed germination of the jujube jujube seeds, but the osmotic effect was dominant. Pomacea jujube seeds show more salt tolerance because of its ability to resist osmotic stress.
