目的探讨干细胞因子(stem cell factor,SCF)对活动低下逼尿肌中Cajal间质细胞(interstitial cells of Cajal,ICCs)数及逼尿肌收缩力的影响。方法将SD大鼠分为3组:对照组、逼尿肌活动低下(detrusor underactive,DU)组以及DU+SCF组。采用ELISA检测血清SCF水平,免疫荧光组织化学法检测ICCs细胞数目,RT-PCR以及Western blot检测c-kit以及SCF的组织水平表达;体外肌条实验检测逼尿肌收缩能力。结果活动低下逼尿肌组织中ICCs数显著减少(P<0.05),SCF及c-kit表达显著降低(P<0.05),逼尿肌收缩频率及收缩幅度显著下降(P<0.05);外源性SCF可以显著增加活动低下逼尿肌组织中ICCs数(P<0.05),增加SCF及c-kit的表达(P<0.05),显著提升逼尿肌收缩幅度和收缩频率(P<0.05)。结论外源性SCF可以影响活动低下逼尿肌组织中ICCs数,进而改变逼尿肌收缩能力。
Objective To investigate the effect of stem cell factor (SCF) on the number of Cajal interstitial cells of Cajal (ICCs) and detrusor contractility in detrusor muscle with low activity. Methods SD rats were divided into 3 groups: control group, detrusor underactive (DU) group and DU + SCF group. Serum levels of SCF were detected by ELISA. The number of ICCs was detected by immunofluorescence staining. The expression of c-kit and SCF was detected by RT-PCR and Western blot. The contractility of detrusor muscle was detected by in vitro muscle strips. Results The number of ICCs in detrusor tissue was significantly decreased (P <0.05), the expression of SCF and c-kit was significantly decreased (P <0.05), and the frequency and amplitude of contraction of detrusor muscle were significantly decreased (P <0.05) Sex SCF significantly increased the number of ICCs (P <0.05), increased the expression of SCF and c-kit (P <0.05), and significantly enhanced the contractile amplitude and contractility of detrusor muscle (P <0.05). Conclusions Exogenous SCF can affect the number of ICCs in detrusor tissue with low activity, and then change the detrusor contractility.
