针对番茄4个抗病基因Tm-2、Pto、Sw-5和Ve1,根据其序列的碱基差异设计引物,经过引物特异性检测和PCR产物克隆测序比对之后,采用高分辨率熔解曲线(High resolution melting,HRM)技术进行多态性检测,共开发了5个SNP标记。经过验证,所开发的标记均能将抗病基因型不同的番茄材料分型,并且分型结果与已知材料的抗病性状完全一致。这些标记可以作为功能标记在番茄抗病育种中应用。
According to the nucleotide differences of their sequences, primers were designed according to the four resistance genes Tm-2, Pto, Sw-5 and Ve1 of tomato. After verification, the developed markers can allotype tomato materials with different disease resistance genotypes, and the typing results are completely consistent with the disease resistance traits of the known materials.
