以磺胺二甲基嘧啶(SM2)作为亲和配基,构建聚乙二醇(PEG)/柠檬酸钠亲和双水相体系,并对β-hCG单克隆抗体在该体系中的分配效果进行研究。结果表明β-hCG单克隆抗体在PEG/柠檬酸钠双水相亲和体系中具有较好的分配特性,进一步考察了成相聚合物分子质量、体系各组分浓度、PEG-SM2加入量、pH值、抗体加入量等因素对分配效果的影响,优化后的分配条件为:PEG 4 000 9%,PEG-SM2 8%,柠檬酸钠浓度12%,pH6.0,抗体加入量0.5 mg/g,抗β-hCG单克隆抗体的分配系数达7.6,收率为91.1%。
